Investigating the Role of CCL20 on IMQ-Induced Psoriatic Inflammation Using CCL20 Deficient Mice.
Psoriasis is an autoimmune inflammatory disease that mainly causes plaques, thick and scaly patches on the skin. Imiquimod (IMQ), a ligand of Toll-like receptor 7/8, has been used for the induction of psoriatic inflammation in mouse models. IMQ enhances keratinocyte hyperproliferation and immune cell infiltration such as dendritic cells and IL-17A-producing T cells. Recently, we have demonstrated that impaired TLR signaling in keratinocytes resulted in the suppression of IMQ-induced skin inflammation with reduced amount of CCL20, a CC-chemokine, in the skin. However, the critical role of CCL20 in psoriasis has not yet been demonstrated using CCL20 KO mice. In this study, IMQ was applied daily on the ear skin of wild-type (WT), CCL20- and CCR6-deficient (KO) C57BL/6 mice for 10 days. The IMQ-treated CCL20KO and CCR6KO mice showed reduced ear thickness, scaling, and erythema compared to the IMQ-treated WT mice. IMQ-treated ear skin showed increased epidermal thickness and subcutaneous tissue in WT mice, while these were significantly suppressed in the mutant mice. RT-qPCR demonstrated reduced mRNA expression of IL-17A and IL-22, but not p40 (subunits of IL-12 and IL-23), in the ears of CCL20KO and CCR6KO mice treated with IMQ. Moreover, IMQ treatment increased the frequencies of γδ+ T cells in both the epidermis and dermis of WT mice but not in the mutant mice. Taken together, these data suggest that CCL20 might be required for IL-17A and IL-22 production as well as γδ+ T cell infiltration in psoriatic inflammation after IMQ treatment. Thus, CCL20 might be a critical molecular target for novel therapies for psoriasis.
