Nanさんの学位論文審査会が開かれました(8月13日)

 8月13日、Nanさんの学位論文審査会が開催されました。審査は、主査の羽多野教授、副査の平野教授、清水准教授により行われ、審査会は英語で実施されました。
 学位論文のタイトルは、“The CCL20-CCR6 axis predominantly drives IL-17-mediated psoriasis while modestly contributing to papain-induced type 2 inflammation” です。
 本研究では、ケモカインCCL20およびその受容体CCR6に着目し、これらを欠損したマウスを用いて、乾癬の病態形成におけるCCL20-CCR6シグナルの役割を解析しました。実験的に乾癬を誘導したところ、CCL20欠損マウス、CCR6欠損マウスのいずれにおいても皮膚炎症が軽減され、CCL20-CCR6シグナルが乾癬の病態形成に重要な役割を果たしていることが示唆されました。
 さらに詳細な解析から、欠損マウスの皮膚病変では、炎症性サイトカインIL-17を産生する病原性γδT細胞の集積が著しく低下していることが明らかになりました。これらの結果から、CCL20-CCR6シグナルを介した病原性γδT細胞の局所への集積が、乾癬の炎症を促進する重要なメカニズムであることが示されました。将来的には、CCL20-CCR6シグナルを標的とすることが、乾癬に対する新たな治療戦略につながる可能性が期待されます。
 審査会では、炎症性皮膚疾患の専門家である皮膚科の羽多野教授、形成外科の清水准教授から、研究内容に関する鋭い質問が相次ぎました。Nanさんは一つひとつの質問に対して丁寧に回答し、これまで積み重ねてきた研究成果と知識をしっかりと示すことができました。  英語での審査という緊張感のある場でしたが、Nanさんにとって、これまでの研究を振り返るとともに、研究者としてさらに成長するための貴重な機会となりました。

Nan’s Doctoral Thesis Defense Held (August 13, 2026)

 On August 13, Nan’s doctoral dissertation defense was held. The examination was conducted in English, with Professor Hatano serving as the chair and Professor Hirano and Associate Professor Shimizu serving as examiners.

The title of Nan’s doctoral dissertation is “The CCL20-CCR6 axis predominantly drives IL-17-mediated psoriasis while modestly contributing to papain-induced type 2 inflammation.”

In this study, Nan investigated the role of the chemokine CCL20 and its receptor CCR6 in the development of psoriasis using mice deficient in either CCL20 or CCR6. When experimental psoriasis was induced, both CCL20-deficient and CCR6-deficient mice showed markedly reduced skin inflammation, suggesting that the CCL20-CCR6 signaling axis plays an important role in the development of psoriasis.

Further analyses revealed that the accumulation of pathogenic γδ T cells producing the inflammatory cytokine IL-17 was substantially reduced in the skin lesions of the mutant mice. These findings suggest that CCL20-CCR6 signaling promotes psoriasis-associated inflammation by facilitating the recruitment of pathogenic IL-17-producing γδ T cells to the skin. Targeting the CCL20-CCR6 signaling pathway may therefore represent a potential new therapeutic strategy for psoriasis in the future.

During the dissertation defense, Professor Hatano, an expert in inflammatory skin diseases, and Associate Professor Shimizu from the Department of Plastic and Reconstructive Surgery posed a number of challenging questions regarding the study. Nan responded carefully and appropriately to each question, demonstrating a solid understanding of the research and the knowledge she has developed through her doctoral studies.

Although the defense was conducted in English and presented a challenging setting, it was a valuable opportunity for Nan to reflect on her research achievements and further develop as a researcher.
